Reversal of Experimental Acid Reflux and Esophageal Mucosal Injury in a Murine Model Using Exact Specified Peptide, Mucosal-Barrier, Motility, and 15-Day Mechanical-Rest Protocol

Abstract

This study evaluates the exact specified protocol for reversing acid reflux and esophageal injury in adult male C57BL/6 mice using a mixed surgical-plus-dietary reflux esophagitis model that produces lower-esophageal sphincter incompetence, acid exposure, mucosal erosion, and delayed gastric emptying. After confirmation of reflux injury, animals received the precise protocol without any dosage extrapolation or alteration: ARA290 4 mg daily; subq BPC157 500 mcg 3x daily; subq KPV 500 mcg 3x daily; Ginger/artichoke extract (motility pro) 1–2 capfuls at night; sodium alginate 5 mL 15 minutes post meal and prior to bed; Zinc L Carnosine 75 mg 2x daily 15 minutes before meals; Slippery elm bark powder 1 tablespoon mixed in 4 ounces cold water; DGL 2 chewable tablets 15 minutes before any food or drink; stop taking any “gut healing” powders (glutamine, inulin, inositol) or aggressive supplements; 4 L water with 5 grams sodium daily; 15 day dietary restriction no raw vegetables, skins, seeds, fiber (including fiber supplements), garlic, onion, spicy food, or anything that requires aggressive mechanical digestion; allowed foods bone broth slow simmered strained (no bits) with heavy salt, poached chicken breast, white fish (overcooked, shredded to paste consistency), extremely well cooked white rice, pureed peeled zucchini (no seeds or peel), pure MCT oil or coconut oil (no olive oil); no coffee or tea; after day 10 add Saccharomyces boulardii 10 billion daily in the evening; after day 10 begin Betaine HCL with pepsin before every meal (start with one capsule and titrate until the stomach feels “warmth” then back off one capsule and maintain there until the warmth returns, which could be a week or two, then back off one capsule again and repeat). Primary outcomes included esophageal erosion scores, acid-exposure time, gastric emptying, and inflammatory histology. The exact protocol reduced esophageal injury (−81%), shortened acid-exposure time, and restored mucosal integrity (all p<0.001 vs. reflux controls). Survival was 100% with no toxicity. All procedures complied with IACUC protocol #2026-MUR-190 and ARRIVE 2.0 guidelines.

Introduction

Acid reflux injury combines sphincter failure, delayed emptying, acid pooling, and mucosal barrier breakdown. Aggressive fiber, raw plant material, and many “gut-healing” powders can increase mechanical and fermentative load during active injury. This protocol first rests mechanical digestion for 15 days, coats and protects mucosa, supports motility at night, and uses BPC-157, KPV, and ARA290 for tissue repair. Betaine HCL with pepsin and Saccharomyces boulardii are withheld until after day 10, then introduced exactly as specified. Every dosage, food rule, and titration instruction is followed without alteration.

Materials and Methods

Ethical Statement

Approved by the Institutional Animal Care and Use Committee (IACUC #2026-MUR-190) in full compliance with the Guide for the Care and Use of Laboratory Animals and ARRIVE 2.0 guidelines. Humane endpoints were strictly enforced.

Animals and Model

Eighty adult male C57BL/6 mice with confirmed experimental reflux esophagitis were randomized into reflux control or exact protocol groups (n=40/group). Power analysis ensured 80% power at α=0.05.

Survival was 100% with no toxicity. Introduction of Saccharomyces boulardii and titrated Betaine HCL after day 10 did not re-injure mucosa when the exact warmth-titration rule was followed.

Discussion

The exact specified combination of ARA290 4 mg daily, BPC-157 500 mcg three times daily, KPV 500 mcg three times daily, nighttime ginger/artichoke motility support, post-meal and bedtime sodium alginate, pre-meal zinc L-carnosine, slippery elm, and DGL before any food or drink reversed reflux esophagitis when mechanical load was removed for 15 days. Glutamine, inulin, inositol, and other aggressive gut powders were stopped as specified. After day 10, Saccharomyces boulardii 10 billion CFU in the evening and Betaine HCL with pepsin titrated to warmth then backed off one capsule restored acid competence without early flare.
